Another of our core technologies at evocatal involves optimization of protein expression through combinatorial approaches. Our range of production strains and our use of established molecular biology tool-boxes allows us to produce biocatalysts at the commercial scale.

Expression systems

We employ different microorganisms in our business activities, including established strains of Escherichia coli and Bacillus subtilis that have been developed into efficient production organisms through the use of tailored expression systems. Along with the technologies described below (see secretion and chaperones), we are able for example to customize promoter strength or mRNA stability, optimizing those processes for the target protein you want to produce. All of our production strains can be employed to produce industrial amounts of enzyme product.

Secretion

One key element of efficient protein secretion is the choice of a suitable signal peptide, which has to be identified for each specific target protein. evocatal employs more than 800 signal sequences from different microorganisms that can be randomly fused with any target protein. Our ultra-modern HT-screening technology makes the search for just the right signal peptide fast and easy. As a general rule, secretion efficiency can be improved significantly with this technology, even in commercial amounts.

Chaperones

Recombinant expressed proteins often have a tendency to form aggregates. This hinders the correct folding of the desired protein, and leads to low or no yield. Molecular helpers known as chaperones can help prevent aggregation. However, it is impossible to predict exactly what combination of chaperone molecules will achieve the best result for a particular target protein. That means that every attempt must be optimized anew. evocatal uses 73 different chaperones produced by three different microorganisms that can be co-expressed individually or in combination with one another, allowing the subsequent identification of the most efficient production strain. Of course, integrating the chaperone into the customer’s own production strain is also an option. This technology (patent pending) can significantly increase protein yield – in some cases quadrupling it. More
